Using of color Doppler and Power Doppler as predictive factor and their correlation with immunohistochemical markers in canine mast cell tumor

Abstract

Carcinogenesis is a process developed in several phases, the angiogenesis is known as an essential phase for tumor progression and metastasis. The aim of the proposed study is evaluate vascularization of mast cell tumors and correlate it with dog's prognosis. The research will be performed in 20 dogs with mast cell tumor, of any histopathological grade. After diagnosis confirmation of mast cell tumor by fine needle aspirates, dogs will be held in the research and anamnesis and physical exam will be done. The nodules with be measured with pachymeter and the lymph nodes will be punctured to look for metastasis. The assessment of tumor vascularity will be done by ultrasound, using Doppler technology with a 10MHZ linear transducer to evaluate the vascularization with color Doppler and Power Doppler. After surgical removal of the nodules, the histopathological graduation according to Patnaick et al (1984) and immunoistochemical study will be performed with the primary antibodies anti-HIF-1a, anti-fator VIII, c-Kit, MIB-1 and caspase-3. Tumor location, metastasis, ulceration, time evolution of the disease and survival in those dogs will be correlated to the histopathological grade, vascular density demonstrated by Doppler and by the number of positive vessels for VIII factor. Datas as imunoexpression to HIF-1 will be correlated with vascular density in Doppler and immunohistochemical study.
