Abstract

The pathology of multiple sclerosis (MS) is typically characterized by perivenular lymphocyte infiltration, destruction of the myelin sheath and axonal loss. The myelin sheath proteins are considered the main targets for T lymphocyte attack. Evidence on the induction and perpetuation of MS still favors CD4+ autoreactive T lymphocytes as a central factor for the autoimmune pathogenesis of MS, in the same way the Regulatory T lymphocytes have emerged as crucial player in the MS. The study of gene expression of different T lymphocytes subsets involved in this disease can determine the possible mechanisms involved in its pathogenesis. Among the different classes of molecules capable of regulating gene expression microRNAs (miRNAs) have aroused great interest in autoimmune diseases. The miRNAs are small RNAs encoded in the human genome that has control function protein expression from the degradation of messenger RNA. Several miRNAs have been described with the regulation of the maturation and differentiation of T lymphocytes and thereby possibly they may be deregulated in MS. Thus the objective of this project is to verify if there is gene expression signature of miRNA in lymphocytes and subsets in healthy patients and with MS in order to better understand the molecular mechanism of this disease.
