Effect of low-level laser and the protein purified from natural latex (Hevea brasiliensis) on the crush injury of the sciatic nerve of rats: Morphological, immunohistochemical and ultrastructural study

Abstract

The treatment of peripheral nerve injury presents itself today as a challenge. New therapies and materials have helped in this context the low-level laser irradiation (LLLI) and natural latex (Hevea brasiliensis) showed positive effects in treating these injuries. The aim of this study is to evaluate the effects of laser and natural latex in crushing injuries of the sciatic nerve (15Kgf, 5.2 MPa, 10 min) of rats. Seventy-two male Wistar rats will be used, weighing 300-400g were divided into six groups (n =12): GI - control, GII - raw nerve; GIII - injured nerve without treatment; GIV - injured nerve and irradiated LLLI (15J/cm2, 780nm); GV - injured nerve may receive the protein purified from natural latex (0.1%) carried by the hyaluronic acid (1% Nikkol) and GVI - injured nerve may receive the protein purified from natural latex (0.1%) adduced by hyaluronic acid (1% Nikkol) and irradiation LLLI (15J/cm2, 780nm). After 4 and 8 weeks of injury, the animals will over-dose of anesthesia and dissected by sectioning the diaphragm for intra-cardiac perfusion. Removal of the left sciatic nerve will be analyzed employing the following methods: 1. Morphologies by light microscopy in cresyl violet staining, Masson and HE 2. Immunoreactivity for NGF, FGF, VEGF and VEGFR-2 and 3. Ultrastructural analysis of tissues with the preparations for transmission electron microscopy and scanning. The results of this study may contribute to the treatment of crush injuries of peripheral nerves (motor and sensory).
