Abstract

Among the drugs used to alleviate or eliminate pain, are the local anesthetics (LA). These compounds are able to block reversibly nerve conduction when applied in a particular region of the body, have relatively short duration of action, but may cause toxic effects on the cardiovascular and central nervous systems. An ideal must AL lasting anesthetic action and toxicity. The dibucaine, a local anesthetic-type amine-amide, has a structure quite different from other local anesthetics in clinical use and, although commonly used in transdermal applications, may have systemic toxicity.Several surveys have been developed in order to prolong the duration of effect and decrease the toxicity of anesthetic compounds. A very promising was opened with the development of sustained release formulations of anesthetic, using carriers that keep the drug for longer at the site of action. One of those ways is to use nanostructured lipid carriers (NLC), produced by mixing lipids solid at room temperature, with liquid lipid (oil) in water.This work aims to prepare nanostructured lipid carriers in order to promote the encapsulation of dibucaine, which will be featured. The formulation produced will be used in in vitro tests to determine their toxicity in cell lines of keratinocytes (HaCaT) and mouse lung fibroblasts (3T3).
