Abstract

Dendritic cell-tumor cell hybrids have been used in therapeutic vaccines for patients with advanced metastatic cancer, achieving promising results. However, both the behavior and the functions performed by these dendritic cell-tumor cell hybrids are still poorly understood, hindering its improvement. In this context, this project aims to study the interaction in vitro of these hybrid cells with different T lymphocyte subsets (CD4+, CD4+FoxP3+, CD4+FoxP3- and CD8+) in a three-dimensional environment. This setting allows cellular migration and the establishment of interactions in a selective way. The project will study also the pattern of lymphocyte responses by analyzing cytokine production by different cells. The main methodology to be employed will be confocal microscopy, and cell-cell interactions, after different time intervals, will be determined by differential staining of lymphocyte subsets. The presence of intracellular cytokines (IFN-gama, TGF-beta, TNF-alpha, IL-2, IL-4, IL-10 and IL-12) in these cells will also be analyzed. Time-lapse microscopy will be used to confirm the kinetics of the major cellular interactions. (AU)
