Effects of leucine and branched-chaim amino acids supplementation on resistance exercise-induced muscle damage and inflammation: a randomized, double-blind, and placebo-controlled study

Abstract

This study aims to evaluate the effects of branched chain amino acids (BCAA) supplementation on resistance exercise-induced muscle damage and inflammation. A cross-sectional, randomized, double-blind, and placebo-controlled study will be conducted. Thirty male, sedentary, and 18-30 years aged subjects will be divided into three groups: BCAA, leucine and placebo (n=10 for each group). Subjects will be randomized according to the isometric strenght. The groups will be submmitted to eccentric exercise protocol consisting of 120 maximal eccentric contractions of the quadriceps muscle performed using an isokinetic dynamometer at angular velocity of 60°/s. The supplementation protocol will consist of 3.5 g of leucine + 3.5 g of alanine for leucine group, 3.5 g of leucine+ 1.7 g of isoleucine + 1.7 g of valine for BCAA group, and 7.0 g of alanine for placebo group. After strength test familiarization, subjects will receive nutritional intervention and consume the supplements during 4 days following the intervention. At baseline, immediately before, after and during 4 days of intervention the volunteers will carry blood collection and assessment of muscle pain. Measurement of isometric strenght will be measured at baseline, after 48 and 96 hours after the exercise session. Additionally, volunteers will undergo muscle biopsies for protein expression analysis and immunohistochemistry at baseline and at the end of the study. Blood samples will be used to measure plasma levels of insulin, glucose, cytokines (IL-1², IL-4, IL-6, IL-10, TNF-±, IFN-³), creatine kinase (CK), and C-reactive protein; biopsies will be used for protein expression of cytokines described and immunohistochemistry to verify inflammatory infiltrates in the muscle fibers. With this, we intend to assess the effectiveness of BCAA and leucine supplementation in attenuating exercise-induced acute muscle damage and inflammation and the possible cellular mechanisms involved in order to propose a possible nutritional therapy.OuvirLer foneticamenteDicionário - Ver dicionário detalhado (AU)
