Abstract

Objective: The aim of this study was to evaluate if the variation of temperature and pH of sodium hypochlorite (NaOCl), increase its antibacterial and dissolution ability on dentin infected in situ. It was also studied if the effect of chitosan nanoparticles (CN) as a final irrigant inhibits the bacterial growth. Material and Methods: The antibacterial and dissolution effects will be studied using the Confocal Laser Scanning Microscopy (CLSM) and Portable Scanning Electronic Microscopy (PSEM) respectively. Two hundred and sixty bovine dentin blocks will be utilized. The samples will be fixed on intraoral device and then a volunteer will used it for 48 hours. Then, the samples will be incubated in BHI for 24 hours to standardize the bacterial growth. The experimental solutions will be 1% and 2.5% at room temperature and 37ºC and acidified at pH 5 e 7. The exposure time will be 5 and 20 minutes. Four operative fields will be evaluated. The specimens will be analyzed pre- (control) and post- irrigation using CLSM. After the analyze procedures, the samples will be incubated in BHI for 24 hours and analyzed again to evaluated the bacterial revitalization. The procedures will be performed in duplicate to determinate the percentage of dentin clean using the PSEM areas. The samples will be irrigated with a solution of CN and then they will be infected intraorally to evaluate the CN antibacterial effect. After the infection period, the samples will be analyzed using the CLSM. For each group will be separated 2 blocks to control procedures. The data obtained will be transferred to the GraphPad Prism in order to perform the statistical analyze. The Kappa test will be used to determinate the concordance inter and intra evaluators. The Kriskal-Wallis Dunn test will be used to perform multiples comparisons among the groups. The significant level will be fixed at P < 0.05.Key words: Sodium Hypochlorite. Dentin. Biofilm.
