Abstract

Objective: the aim of this study will be evaluate the efficacy of chitosan nanoparticles to inactivate bacteria and prevent biofilm formation at the sealer-dentin interfaces. Material and methods: The study will be divided in three stages. First stage: the experiment will be conducted to analyse the antibacterial properties of carboxymethyl-chitosan (CMCS) in different formulations against biofilms. Second stage: direct-contact and membrane-restricted methods will be utilized to evaluate the antibacterial ability of an epoxy resin (ThermaSeal Plus) (TS) and calcium silicate (MTA Fillapex) (MTA) based-sealers associated or not with chitosan nanoparticles (CNp). Third stage: the biofilm formation at the sealer dentin interfaces of root canal dentin treated with CMCS and filled with gutta-percha and sealer incorporated with CNp will be analyzed after 1- and 4- weeks aging periods. The samples will be treated and filled as follow: (1) distilled water - unaltered sealer (control group), (2) CMCS - sealer+CNp (CMCS group), (3) CMCS/Rose Bengal - sealer+CNp (CMCS/RB group). Microbiological and microscopic analyses will be used to perform the study. E. faecalis will be used to infect the samples throughout the experiment. (AU)
