Abstract

Although conventional endodontic treatment significantly reduces the microbiota present within the root canals, the permanence of microorganisms due to the anatomical complexity of the root canal system and their resistance to chemical-mechanical treatment can lead to persistent or secondary infections. In addition, the irrigating agents most used in Endodontics have shown cytotoxicity and genotoxicity, especially in the higher concentrations, being toxic to the periapical tissues. All these effects are complicated in the case of immature permanent teeth, since the wide opening of the apical foramen further promotes contact of these products with the residual vital tissue responsible for the complete formation of the root. Many authors have explored the use of phytochemicals, such as curcumin and cinnamaldehyde, in order to find biological compounds that have antimicrobial properties. The synthesis of curcumin hybrids with another agent with similar antibacterial potency, such as cinnamaldehyde, would be an interesting strategy to increase its therapeutic potential and reduce the limitations of curcumin alone, such as low solubility and staining. This study aims to evaluate the cytotoxic effect and antimicrobial action/antibiofilm of hybrids of curcumin and cinnamaldehyde on microorganisms of endodontic interest. Curcumin, cinnamaldehyde and curcumin-cinnamaldehyde hybrid compounds will be evaluated. The antimicrobial activity of the compounds will be evaluated by the Minimum Inhibitory Concentration (MIC) and Minimum Bactericidal Concentration (CBM) assay on Streptococcus mutans, Lactobacillus acidophilus, Actinomyces israelii, Enterococcus faecalis and Fusobacterium nucleatum. The effect of the compounds on the biofilm will be verified by means of Of the evaluation of the metabolic activity of the biofilm by colorimetric method (XTT) and also by the count of CFU/ml. The assays will be conducted with isolated biofilms and dual-species. It will also be evaluated the effect of the compounds on the biofilm formed in bovine root dentin by analysis in Confocal Microscopy. Finally, the toxicity of the compounds on fibroblasts (L-929) will be investigated by the MTT method. All experiments will be performed in triplicates and on independent days. The data will be analyzed statistically. In the case of data presenting normal distribution and homogeneity of variances, the variables can be submitted to analysis of variance (ANOVA) followed by the Tukey test, if they do not meet the assumptions, will be submitted to non-parametric tests, such as Kruskal-Wallis and Mann- Whitney. All analyzes will be performed considering a significance level of 5%. (AU)
