Abstract

The aroeira's extract study and their effects in specific tissues and cells can contribute significantly to the understanding and validation of your property like herbal. The aim of this study is to evaluate the influence of aroeira's extract in human gingival fibroblast and the matrix metalloproteinase 2 and 9 activity. First, the fibroblasts will be plated with media Eagle modified by Dullbecco complement for 10% fetal bovine serum. After the fibroblasts adhesion, DMEM will be substituted in a culture medium with different aroeira hydroalcoholic extract concentrations (pure extract; 1:10; 1:100; 1:1000; 1:1000 and control). After 24, 48, 72, 96 and 120 hours of the treatment with the different extract concentration, the sample will be collected, shared, and analyzed. The collected samples will be available the cellular viability by cells mitochondrial activity test will be realized by the MTT's reduction method (bromate 3-(4,5-dimetiltiazol-2-yl)-2,5-difeniltetrazólio), neutral red capture and violet crystal. The other sample will be used to available the MMP 2 and 9 activity for zymography. The values will be evaluated by ANOVA's method, complemented by Dunn's test when submit to normal or abnormal distribution, respective. The significant level used will be 5% (p<0.05).(AU)
