Abstract

Due to the indiscriminate use of antibiotics, the resistance of pathogenic microorganisms to various drugs has increased in recent years. This situation has forced researchers to search for new drugs. An alternative is the herbal medicines that are pharmaceutical preparations (extracts, tinctures, ointments and capsules) of medicinal herbs, used to treat numerous diseases. The advance is among the plants with antimicrobial and antifungal. One of the Brazilian species of woodland that have shown interest in various areas is the aroeira, by presenting anti-inflammatory action. Osteoblasts are young cells with intense metabolic activity and responsible for producing the organic part of the matrix. The activity of osteoblasts depends on the actions of some modulators such as growth factors such as Bone Morphogenetic Proteins (BMPs), cytokines, matrix metalloproteinases (MMPs), alkaline phosphatase and tartrate-resistant acid phosphatase (TRAP), among others. The aim of this study is evaluate the effect of the extract of aroeira on the proliferation and mineralization of human osteoblasts and whether there is any relationship between the effect of the extract with the expression of MMP-2 and -9; Osteopontin (OPN), bone sialoprotein (BSP), BMP-2 and -7 and Human Ankylosis (ANKH) produced by these cells. For the experiment will be collected aroeira in the campus of UNESP, and you will be extracted from hydroalcoholic extract of aroeira. Then will be cultivated human osteoblasts in ±-MEM medium supplemented with 10% Fetal Bovine Serum (FBS). The experimental groups will be divided into different concentrations of the hydroalcoholic extract of aroeira (pure extract, 1: 10, 1:100, 1:1000, 1:10000). The analysis of cell viability will be done by Uptake of Neutral Red, Reduction of MTT and Crystal Violet, to analyze the expression of MMP-2 and -9, OPN, BSP, BMP-2 and -7, and ANKH through RT- PCR and Western blot. The statistical analyses will be made through the statistical test one-way ANOVA complemented for the test of Tukey, test of Kruskal-Wallis complemented for the test of Dunn and for the "t" test, according to the indication (p <0.05).
