This study aims to evaluate the metalloproteomics profile of samples from Nile tilapia (Oreochromis niloticus) muscle tissue. Two-dimensional polyacrylamide gel electrophoresis (2D-PAGE) will be used for the fractionation of proteins from the muscle tissue samples and for identification of these proteins by analyzing the images obtained by the end of the electrophoretic process. Calcium, Copper, Iron, Manganese and Zinc mapping in protein spots will be made by Synchrotron Radiation X-ray Fluorescence. Different methods for protein extraction and different programs for protein separation by 2D PAGE will be tested. The protein spots containing Ca, Cu, Fe, Mn and Zn will be characterized by ESI-MS. The characterization of these metalloproteins can be a springboard for the Nile tilapia's metallomics. It can also provide important information for the area of fish nutrition and physiology, bringing advances for the proteome and metallome study of this species.
