Expression and localization of EGFR in non-small cell lung cancer: its role in proliferation and cell migration

Abstract

The lung cancer cells frequently have changes in their molecular genetics, for example overexpression of ErbB1 gene. ErbB1 gene encoded EGFR, a receptor tyrosine kinase that is mainly responsible for cell proliferation and survival. EGFR Overexpression has been associated with more aggressive disease, poor prognosis, low survival and low response to therapy. There are many studies focused on the ErbB1 gene amplification, mutations and EGFR overexpression, but there are many controversies in the literature on these aspects. HK2 and A549 cell lines, derived from human lung carcinoma, have been used as models to study in our laboratory. Under these conditions we propose to characterize these two cells, in a 2D and 3D culture system, on the number of copies of gene ErbB1 gene and level of EGFR expression. The activation of EGFR will be inhibited by the tyrosine kinase domain inhibitor AG1478 to evaluate the influence of this receptor on the proliferation and cell migration in these cells. (AU)
