Abstract

Rates in vitro produced bovine embryos is related to efficiency of IVM procedure that intrinsically limits the developmental competence of the oocyte. Thus , by determining the mRNA expression of Cx43 , GDF -9 , BMP15 , OOSP1 and fibronectin in oocytes and GREM1 , PTGS2 , PFKP , VCAN , HAS2 and AREG genes in cumulus cells , this study was designed in order to adapt a culture system that enhances in vitro maturation, especially cytoplasmic causing an increase in the rate of IVP bovine embryos. Three experiments will be performed to determine gene expression in oocytes matured in vitro under different conditions, comparing with in vivo matured oocytes from superovulated animals: Experiment 1, in media containing different protein sources divided into G1 (10% FCS) , G2 (1 mg / ml PVA) and G3 (4 mg / ml BSA),; Experiment 2, in media containing different concentrations of EGF divided into G1 (0 ng / ml), G2 (10 ng / ml) and G3 ( 100 ng / ml), and Experiment 3,, oocytes undergoing pre-maturation with 10 mM butyrolactone I (BL I) divided into G1 (without prematuration) , G2 (12 hours of prematuration and 12 hours of maturation) and G3 (24 hours of prematuration and 24 hour of maturation). The statistical analysis will be complemented by Kruskal-Wallis Mann-Whitney. Differences of p <0.05 are considered as significant. (AU)
