Refrigeration and freezing of canine semen diluted in Tris-egg yolk containing antioxidant

Abstract

The present study will be conducted to evaluate the effect of adding different concentrations of antioxidants, vitamin E analog (Trolox C), to the canine semen that will be submitted to cooling and freezing. Six collections will be held from 12 Beagle dogs, of which a semen pool will be originated of 3 animals ejaculated. First, will be done an evaluation of the physical characteristics by observing the volume, color, and viscosity of the semen pools, which will be determined spermatic motility, spermatic vigor, and spermatic concentration. After the evaluation of the physical and morphological characteristics of the ejaculates, each pool will be divided into samples, which will be added to the Tris-egg yolk extender (control) and increased by 0,1 and 0,2 mMol of antioxidant and will be submitted to lipid peroxidation test (TBARS), fluorescence test and total antioxidant capacity (TAC) for then the samples are chilled at 4 R C (for 48 hours and evaluated) and frozen. Then, sperm morphology and additional tests will be repeated with each extender after cooling and freezing the samples. The results will be processed through the SAS system and analyzed by general linear models, analysis of variance (ANOVA) for repeated measurements, using the cooling and freezing like effect intra-group and the type of extender as inter-group effect (fixed effects). The levels of repeated measurements will be n = 6, for the determinations after the cooling (48 hours) and the freezing of straight progressive motility and additional tests. Statistical analysis will be performed by the SAS system, which will be tested the analysis of variance and then will be held the comparison of means by Tukey's test, being the differences considered significant at probability P <0.05.(AU)
