Abstract Iron overload may induce a great diversity of tissue injuries, mainly due to the onset of labile plasma iron (LPI) reservoirs, which include the redox-active species of this metal in the organism. Ideal ligands to iron chelation therapy should be able to penetrate iron-overloaded tissue, coordinate the metal in stable and redox-inactive forms, remove it from the cell and transfer it to the circulatory transferrin or other siderophores. In this project, we propose the study of isatin-Schiff bases and phytate as depletors of LPI in serum samples of iron-overloaded patients (thalassemia, hemochromatosis, bone marrow transplant). Also, their relative affinities towards iron in physiological medium will be probed by calcein and fluorescent transferrin. Finally, the abilities of these chelators to decrease intracellular pools of labile iron will be investigated.(AU) | |
