Abstract

A presence of microcystin into water can be fatal to wildlife and domestic animals and results in various diseases in humans. This project aims to determine the susceptibility of the trairão, Hoplias lacerdae, to microcystin present in the crude extract of M. aeruginosa with emphasis on its effects and mechanism of action in liver and muscle of this species and its bioaccumulation as potential risk for human health. Two experimental protocols will be conducted to test the toxicity of microcystin. In the first protocol, juvenile of H. lacerdae will receive intraperitoneally the crude extract of microcystin and after 6, 12, 24, 48 and 96 hours, the plasma, liver and muscle will be sampled and analyzed to evaluate the response time after acute contamination. In the second protocol, the animals will be chronically contamined, every 96 h, for 32 days and the changes in the liver and muscle will be evaluated as well as the toxin bioaccumulation in these tissues. The toxin in the tissues will be identified by inhibition of protein phosphatases (PP1 and PP2A) and the confirmation and quantification of microcystins in the tissues will be done by HPLC-PDA. Its location will be done using immunohistochemistry. Changes in liver and muscle tissue will be evaluated by biochemical analysis of plasma (activity of alanine aminotransferase, aspartate aminotrasferase, phospholipase A2 and concentration of bilirubin and tumor necrosis factor-±); plasma, liver and muscle (acid and alkaline phosphatase, intermediary metabolites); liver and muscle (biomarkers of oxidative stress). Morphological studies will be performed to determine the degree of liver injury using light microscopy techniques (histopathology, immunostaining of apoptotic cells and cell proliferation) and transmission microscopy.
