Abstract

Semen cryopreservation is an important technique that has been improved last years impelled by aquaculture development. The benefits of sperm cryopreservation include synchronization of gamete availability of both sexes, sperm economy, simplification of broodstock management, transport of gametes from different fish farms, and germplasm storage for genetic selection programs or conservation of species. Nowadays there are two semen cryopreservation methods: conventional congelation(slow freezing) and vitrification (ultra fast freezing) that differ at concentration of cryoprotectants, and the cooling and warming rates. The purpose of this study is to determine different ways of cryopreservation of Rhamdia Quelen (vitrification/ conventional congelation) and estimate the seminal quality and fertilization aptitude before and after cryopreservation. It will be evaluated the seminal morfofuncional characteristics pre and post thawed, toxicity of three cryoprotectants (Metanol, DMSO, Metil glicol) followed by conventional congelation with the best diluents observed in the toxicity of cryoprotectants. Afterward it will be carry out the vitrification of the Rhamdia quelen semen, with and without cryoprotectants.
