Abstract

The induced pluripotent cells (iPS) were first reported in 2006 and has been extensively studied. Through the technique, adult somatic cells acquire behavior very similar to embryonic stem cells, reducing the ethical issues related to the use of such research. However, the biological mechanisms of iPS cells are not yet fully elucidated. Thus, further research is needed to ensure the safety and effectiveness of their possible use in future therapies. Adipose derived stem cells consists of a type of multipotent mesenchymal stem cell that is highlighted by the ease, speed and security of collection and processing. The rabbit iPS cells from adipose derived stem cells will be produced using the introduction of four transcription factors (Oct3/4, Sox2, c-Myc, and KLF4). The behavior of the cells will be analyzed for pluripotency markers Oct4 and Nanog, alkaline phosphatase activity, ability to differentiate into embryoid bodies in vitro and tumor formation in vivo. From these data will be carried out comparative analyzes of adipose derived stem cells and iPS cells of rabbits. From these data, comparative analysis will be performed between adipose derived stem cells and iPS cells of rabbits. The aim of this study is to establish an adequate protocol to produce iPS cells from adipose derived stem cells of rabbits. We expect to develop a Brazilian technology still unpublished in rabbits, and to create a lineage of iPS cells for future application in cellular therapy.
