Abstract In the past decades many advances have been made on the study of the Renin Angiotensin System (RAS), with the identification of new components and the consolidation of the existence of an intracellular system with independent modulation from the systemic RAS. It is believed that the intracellular RAS is activated in particular pathological situations, for instance under hyperglycemia.Thus, in view of the results obtained so far with the master's project entitled "Pathophysiological role of the Angiotensin Converting Enzyme 2 (ACE 2) in kidney and heart of an animal model of sepsis" (FAPESP Process 2011/04940-2) in which we see that the E. coli lipopolysaccharide (LPS) is capable of influencing the tissue RAS, it would be interesting to add it a new level of complexity and study the direct effects of the LPS treatment on the intracellular RAS. To do so, we would use primary cultures from renal and cardiac cells.Our laboratory already has experience in mesangial cells primary culture, but we have no experience in cardiomyocytes culture, this technique is often used by the group of Professor Rajesh Kumar (Cardiovascular Research Institute, Texas A & M HSC, College of Medicine, USA).Therefore, in this project, we propose an internship of the student Rodolfo Mattar Rosa in the laboratory of Professor Dr. Rajesh Kumar in order to learn techniques of cardiomyocytes and cardiac fibroblasts cultures and methods of analysis of the intracellular RAS, for implementation and dissemination in future projects of our research group.
