Abstract The effects of the 470 nm wavelength (blue light) produced by lighting emitting diodes (LED) on odontoblast-like cells have yet to be elucidated particularly regarding the production of components of the dentin matrix which are essential for pulp repair. Considering that the blue light is already used in clinical procedures to photo-activate liner/restorative materials, it would be interesting to demonstrate if that same wavelength is capable of inducing pulp repair. If that is true, the data from the present study would support further investigations regarding to the development of a prototype that could, at the same time, be used in restorative procedures and also, after the necessary adjustments such as irradiance and energy dose, be used to stimulate pulpal cells in order to promote the repair of the pulp tissue. Thus, the purpose of the present study will be to evaluate the effect of photobiomodulation using a 455 nm LED on the production of dentin matrix-related proteins by odontoblast like-cells MDPC-23. The cells will be seeded in acrylic culture plates and submitted to nutritional deficiency by reducing the percentage of bovine fetal serum in the culture medium. Then the cells will be irradiated a single time with different doses of energy: 0.5, 2, 4, 12 or 15 J/cm2. After 72 hours, it will be determined the cell viability (MTT), mineralization nodules (Alizarin Red), total protein (Read Northcote), alkaline phosphatase activity and collagen production (Sircol Red). Statistical tests will be selected based on the type of response variable and data set properties. All statistical inferences will be taken at the 5% level of significance. | |
