Abstract

The proposal objectives are: Experiment I - Study the effect of subspecies (Bos taurus indicus vs. Bos taurus taurus), different embryo culture media (SOFaa + 2.5% FCS versus SOFaa + 2.5% FCS + 0.3 µM PES) and embryo condition (fresh versus vitrified) on global gene expression pattern (microarray) cryotolerance, lipid content (Sudan Black), and lipid fingerprint mass spectrometry profile (MALDI-TOF MS) of in vitro produced embryos (OPU-PIV). Experiment II - Evaluate the effect of different embryo culture media (SOFaa without FCS; SOFaa + 2.5% FCS; SOFaa + 2.5% FCS + 0.3 µM PES, and SOFaa + 10% FCS) and embryo condition (fresh versus vitrified) on embryo development, lipid content, lipid fingerprint mass spectrometry profile, cryotolerance, and relative gene expression pattern of in vitro produced embryos (PIV). Experiment III - Analyse the effect of subspecies (Bos taurus indicus vs. Bos taurus taurus) and embryo condition (fresh and vitrified) on global gene expression pattern, cryotolerance, lipid content and lipid fingerprint mass spectrometry profile of in vivo produced embryos (SOV).
