Abstract

Koinobiont parasitoids manipulate their host growth and development by altering the host endocrine system, gene expression and metabolism to adequate the host environment to suit the parasitoid own needs. Regulation of the host gene expression and metabolism are key processes to assure a successful parasitization, e.g host colonization and utilization by the immature parasitoid. THerefore, this project aims to evaluate the effect of the parasitization by Campoletis flavicincta on the gene expression and protein sysnthesis of larvae of Spodoptera frugiperda at different stages of parasitoid development. The effects of parasitism on the host will be evaluated by comparative transcriptomic analysis using the new generation sequencing technologies (NGS) via RNA-Seq analysis in an Illumina platform. THe initial information on the differential gene expression obtained will be confirmed by quantitative PCR (qPCR). Changes in the protein profile of the host due to parasitization will be investigated by SDS-PAGE and MALDI-TOF analyses. Due to experimental conditions, RNA-Seq analysis will focus on the initial stages of parasitization (host colonization - 06, 24 and 73 h after parasitism), while the protein analysis will be cover the entire period of parasitoid development (1, 3, 5, 7 and 9 days after parasitization).
