Abstract

Nanotextured titanium-based surfaces promote enhancedosseo integration since they influence important cellular events, such as the synthesis of relevant biological molecules including osteopontin (OPN). Recent studies approach the importance of OPN in macrophage differentiation, migration, adhesion and production of proinflammatory mediators. Macrophages have a major role inhost inflammatory and immune processes; they are a major source of cytokines, chemokines, and growth factors involved in early stages of wound healing after implant insertion. Therefore, their response to biomaterials is extremely important in understanding material-mediated host response. In the present study, we aim to better understand the macrophages behavior concerning OPN expression when they are cultured on a Titanium (Ti) nanotextured surface. The Raw 264.7 macrophage cell line will be cultured on Ti discs with nanotextured or machined surfaces. The expression of OPN protein will be evaluated by indirect immunofluorescence and the gene quantification will be verified by Real-Time PCR. The analyses will be performed at two experimental times.
