Abstract

Topiramate is one of the drugs recommended for treatment of epilepsy, with multiple mechanisms of action, which also has demonstrated pharmacological potential for the treatment of obesity. Pharmacological tests regarding the side effects of Topiramate on reproduction include mainly its effects on fertility and embryo fetal toxicity, and there are very few studies in these aspects. In addition, none report was obtained about the effects of the drug in ovarian structure and if there are evidences of fetal alterations that may occur due maternal exposition to the drug in the first stages of pregnancy. This knowledge is important, since a woman may not be aware of her condition of pregnancy while using this drug, mainly in the start of gestation.The goal of this study is evaluate the ovarian tissue and the pregnancy outcome of albino female rats exposed to treatment with Topiramate. There will be formed two groups: control - CG (tap water) and treated with Topiramate - TG (100 mg/kg body weight). The treatments will be realized by single dose, daily, during six consecutive weeks (Experiment I), for evaluation of the estrous cycle and histomorphometry ovarian. In Experiment II, pregnant rats of groups CG and TG will receive the treatments in two different periods of pregnancy: pre-implantation (days 1 to 4) and implantation until the start of organogenesis (days 4 to 7), to the evaluation of reproductive and fetal parameters.(AU)
