Abstract

The cell signaling is the mechanism that cells have to communicate with their neighbors, distant tissues or even in internal processes. In leukemia, disorders are often detected in multiple signaling pathways, making it vital to seek knowledge of these pathways and new therapeutic targets. It has been reported that the protein SIVA1 participates in signaling through p53 tumor suppressor and recent in vivo experimental data related their function the oligomeric structure. Therefore, the question arises which the structural characteristics of the oligomers formed by SIVA1 and inhibition of this process could lead to activation of the p53 tumor suppressor in the treatment of hematopoietic disorders. The purpose of this project is therefore to characterize the structure of the protein SIVA1 by crystallography and nuclear magnetic resonance, two complementary methods in macromolecular structural analysis. The SIVA1 protein will be hyperexpressed in a large scale and purified. Data collection for the study of the structure will be made at the National Laboratory Biosciences (LNBio) by submitting research proposals. Other biophysical techniques will be used to complement the structural information obtained, which will be analyzed in the Molecular and Cellular Biology Laboratory of Hematology Center at UNICAMP and also in the Molecular Biology and Circular Dichroism Laboratory of Chemistry Institute at UNICAMP. All this information will bring great contribution to scientific literature, and will assist in understanding some mechanisms of cell signaling pathways in hematologic malignancies. (AU)
