Abstract

The establishment and maintenance of maternal tolerance to fetal antigens are essential for successful pregnancy in animals Eutheria. The gamma delta T cells (³´T) are present in the endometrium / decidua during pregnancy in humans and mice. The amount of this gamma delta T cells in the uterus appears to increase with advancing gestation. The objectives of this project are to identify the presence of ³´T cells in the uterine endometrium in pregnant and non-pregnant cows; evaluate the population dynamics of ³´T cells in peripheral blood in early (around day 33 after insemination) and in the peripartum period cytometric flow, and analyzing cytokine expression profile of purified samples of ³´T cells by flow cytometry by means of real time quantitative PCR comparing cells from peripheral blood of pregnant cows with samples of cells from non-pregnant cows.The hypothesis of this project that there is a ³´T cell recruitment to the endometrium throughout gestation and express cytokines that promote the establishment and maintenance of maternal tolerance to fetal antigens during pregnancy in cattle.
