Abstract

The genetic structure of natural populations can reveal interesting patterns regarding the evolution and early stages of speciation. The extensive amount of molecular markers available offers an interesting opportunity to investigate the evolution of early stages of speciation, mainly in natural populations. The main objective of this study is to apply the AFLP (amplified fragment length polymorphisms) technique to analyze the genetic differentiation patterns of natural populations of Epidendrum secundum (Orchidaceae), including its geographic distribution along Central Brazilian Range and Atlantic Forest. Approximately 350 individuals sampled on 16 populations will be analyzed using ten different primer combinations of AFLP markers. Bayesian assignment analysis will be applied in order to identify major genetic clusters. Gene flow patterns will be estimated in order to investigate differential gene flow among loci, using the software INTROGRESS. The early stages of speciation and the existence of incipient species will be investigated using the plastid sequences dataset which will be available, as described in the main postdoc project. (AU)
