Interaction between galectin-1 and integrin during adhesion and migration in mast cells

Abstract

Galectins are a family of lectins which share similar carbohydrate recognition domains which is responsible for ²-galactoside binding (CRDs). Galectin-1 is present both inside and outside cells, however secreted form have higher lectin activity. Galectin-1 modulates the cell adhesion and spreading, but this function depends on the cell type. Mast cells present adhesion molecules on surface for to do its functions. Studies show that the substrate adhesion is an important regulatory factor in the mast cell secretion and on surface, which mediate the interactions cell to cell and cell to substrate. Our results showed that RBL-2H3 mast cell line express galectin-1. Also, these results demonstrated that deficient mast cell line in gangliosides of type GM1 and GD1b (D1 cells) do not express galectin-1. D1 cells present structural and functional alteration. This study revealed that transfected D1 cell with galectin-1 induces occur a structural reorganization that reflects in the cell spreading on the substrate. In order this evidences the present study has aims to investigate the molecular mechanism whereby the galectin-1 act in the mast cell spreading, adhesion and migration. For this, in addition to D1 cells RNA interference will be performed in RBL-2H3 cells. The different integrin subunits and proteins related to cell adhesion like as FAK and vimentin will be investigated during the process of cell adhesion and migration. The known of the molecular mechanisms whereby the galectin-1 can to act in the adhesion and migration of mast cells will contribute for the mast cells biology understanding in the allergic and inflammatory processes.
