Abstract

Mangabeira (Hancornia speciose) is a Brazilian native fruit tree that can be found mostly on the northeast coast of Brazil. The latex and some other parts from the Mangabeira tree have been popularly used as an auxiliary treatment for bone fractures reporting benefits. In dentistry, bone support is necessary both for the attachment of the tooth in the bone and for prosthetic rehabilitation. Therefore, the stimulation of bone formation is needed in several situations. Based on the popular usage of latex as an aid to the treatment of bone fractures, some pilot studies were performed in our laboratory to verify the response of the bone tissue to the latex of Hancornia speciosa. The results achieved so far indicate the latex potential to induce bone formation. Thereby, we decide to investigate if the latex is stimulating progenitor cells to differentiate into mature osteoblasts and/or if the latex could lead to bone formation by stimulating the proliferation of osteoprogenitor cells with subsequent differentiation into osteoblasts. In order to study cell proliferation, the gold standard method has been the in vivo labeling of the DNA by 5-bromodeoxyuridine (BrdU), a synthetic thymidine analog. When BrdU is administered in laboratory animals it is incorporated into the nucleus during the DNA synthesis S-phase of the cell cycle and is subsequently detected in histological sections by immunohistochemistry with an anti-BrdU antibody. This project aims to verify that the latex induces the proliferation of osteoprogenitor cells as a requirement for the differentiation into actively synthesizing osteoblasts, using the BrdU labeling method.
