Abstract

Monoclonal antibodies represent an important kind of biopharmaceuticals with wide therapeutic application, especially against cancer. Bevacizumab, a monoclonal antibody that will be used in this study, it acts by preventing the development of angiogenesis factor which accompanies neoplastic tissue and in combination with cytotoxic agents can reduce the tumor size. However, like many other drugs of protein nature, formulations of antibodies represent a big challenge both from a technological standpoint as biopharmaceuticals. The low physicochemical stability in the formulation and biological fluids and low bioavailability and immunogenic potential shows necessary the development of new strategies to overcome these problems. The buccal route, besides having characteristics suitable for administration of proteins, has generated great interest as a route because of its high permeability, high blood flow and absence of first-pass hepatic metabolism. The nanostructured systems with capacity mucoadhesive, like polymeric nanoparticles, also constitute a valuable strategy for mucosal administration of biopharmaceuticals, they increase the bioavailability, protects the three-dimensional structure of the protein and shows low cytotoxicity. This research aims to develop and characterize chitosan-based nanoparticle systems (QS) associated with dextran sulphate (DS) for buccal administration of BCB. The aim is to develop a system for incorporation of this AM in nanostructural level in order to optimize its use in the treatment of oral cancer. (AU)
