Abstract

The impact of excess weight on men´s reproductive health has received increasing attention due to the recent and fast increase in the prevalence of obesity worldwide. According to the Brazilian Institute of Geography and Statistics more than 609 000 Brazilians have class III obesity. In recent decades, many studies have associated the high levels of body mass index (BMI) with the decline in quality functional sperm. Obesity can affect male fertility through three main mechanisms: (i) changes in endocrine profile, (ii) increase scrotal temperature and (iii) a chronic systemic inflammatory process. However, little is known about the post-genomic mechanisms. These are necessary , because studies that address the protein profiles and the interactions that make up the men´s semen in overweight and obesity conditions. Thus, this project proposes to study men without seminal changes and these are divided into three groups according to the difference in BMI, representing control group BMI ³ 18.5 and <25 kg/m², group II with BMI ³ 25 and <30 kg / m² and Group III with BMI ³ 30 kg / m². Then it will perform, firstly, research into the main functional characteristics of sperm, such as mitochondrial activity analysis through staining using DAB (3,3 '-diaminobenzidine); evaluation of acrosomal integrity, using PNA (Peannut Agglutinin) probes and DNA integrity analysis, through the alkaline Comet assay. Analysis of plasma proteomic semen and sperm will be performed using the spectroscopy 2D masses nanoUPLC-ESI-MSE technique. This study hopes to assess the effect of overweight and obesity onsperm function and on semen metabolism. (AU)
