The effects of sleep restriction in an animal model of ischemic brain injury

Abstract

The stroke is a cardiovascular disease of high prevalence and incidence in the world population responsible for different comorbidities that often incapacitate the patients to return to their daily activities. Sleep quality has been identified as an important predictor or preconditioning factor for the occurrence of ischemic stroke. Thus, the aim of this study is to evaluate the effects of sleep restriction (SR) in rats subjected to transient middle cerebral artery occlusion (TMCAO), considered an experimental model of ischemic stroke. We will investigate the mortality rate, the neurological function and the ischemic lesions severity. Male 2 months-old Wistar rats will be utilized. The animals will be submitted to the SR protocol for 18 hours during 21 days previously to TMCAO protocol. The severity of ischemic lesions will be evaluated by the mortality rate, cellular proliferation and death rate, the infarct volume and the neurological function after 7 days of stroke recovery. We hope with the present project perform a translational approach that allows to better understand the role of sleep and its effects on recovery of ischemic lesions in stroke patients.
