EFFECTS OF EXENATIDE AND SITAGLIPTIN ON INDUCED PERIODONTITIS IN RATS

Abstract

Diabetes Mellitus is a high prevalent pathology as well as periodontitis. The literature supports a two way relationship between them. New class of drugs for diabetes treatment, peptide-like glucagon-1 (GLP-1) agonists and dypeptidyl peptidase -4 (DPP-4) inhibitors have been shown pleiotropic effects beside glicemic control, like anabolic effects, inhibition of resorption and antiinflammatory actions. The aim of this study is evaluate the effect of exenatide (GLP-1 agonist) and sitagliptin (DPP-4 inhibitor) on induced periodontitis in rats. Forty male rats will be divided into four groups: (1) exenatide group: treated with 3µg/Kg/day of exenatide subcutaneously, (2) group sitagliptin: treated with 10 mg/Kg/day with sitagliptin orally, and control groups with (3) and without (4) insertion of ligature around lower first molar (treated with the drug vehicle). All animals will be treated for 28 days, when they will be sacrificed and we will extract the gingiva around the first molar to analyse gene expression of SOST, RANK-L, DPP-4, IL-1-beta, COX-2, MMP-9, iNOS, OPG and OC with Real Time (q) PCR. Left mandibles will be descalcified and stained with Hematoxilin-Eosin and Picrossirius Red to analyse bone loss and amount of collagen in gingiva. Bone turnover markers will be analyzed by immunohistochemistry with anti-RANKL and anti-OPG. Quantitative results will be submitted to exploratory analysis to determine the appropriate statistic test that will be applied.
