Viability of sexual transmission of Toxoplasma gondii (Nicolle & Manceaux, 1909) by biotech artificial insemination in female seronegative bovine (Bos taurus x Bos taurus indicus)

Abstract

Toxoplasmosis is a zoonotic disease and cosmopolitan, having importance in human and veterinary medicine, as a promoter of abortion and congenital disease in several species of intermediate hosts. The infection in ruminants occurs by ingestion of oocysts in food and stabling contaminated with the feces of felids, increasing the risk of infection. The aim of this study was to prove the viability of sexual transmission of Toxoplasma gondii in cattle breeding, serologically negative for toxoplasmosis, undergoing artificial insemination with semen experimentally contaminated with tachyzoites this coccidia and cryopreserved according to biotechnologies used in cattle reproduction, indeed unprecedented in the literature. In males assess the maintenance and survival of T. gondii in semen cryopreserved in DMSO and frozen in liquid nitrogen (-196 °C), for the application of biotech fixed time artificial insemination in females and evaluate clinical and laboratory cows of reproductive age, exposed to artificial insemination with cryopreserved semen proven parasitized by T. gondii. Search by indirect fluorescent antibody test (IFAT), the humoral immune response of females post-insemination and after evaluation of all the results obtained, will be inferred from the occurrence or non-sexual transmission, by artificial insemination, T. gondii in cattle. For the values of the clinical parameters of females and males semen will be used completely randomized design, considering the collection of data for each experimental day as replicates and the averages will be compared by Tukey test at 95% confidence. Data from females (clinical parameters) will be analyzed in a factorial split plot the Proc Mixed procedure of the SAS statistical package and the confrontations of the treatments will be obtained by the Tukey-Kramer at 95% confidence.
