Abstract

Peritonitis and its complications have clinical relevance and a potential risk of death to horses, representing a diagnostic and therapeutic challenge. In this sense, new therapeutic possibilities must be tried to increase survival of affected animals. The objective is to evaluate randomized clinical trial by ceftriaxone concentration evolution in peritoneal fluid following intraperitoneal and intravenous administration as well as the feasibility and safety of using Tenckhoff catheter and peritoneal route in horses. The project will be divided into two phases, phase 1: a pilot project to establish the feasibility of using Tenckhoff catheter, ideal dose and ceftriaxone intraperitoneal interval of application to maintain the drug within their minimum plasmatic inhibitory concentration. Phase 2 include the implementation of the project itself, which will be used ten adult horses, in two groups of five animals. Group 1 will receive the ceftriaxone dose established in pilot study diluted in 2 liters of saline solution 0.9% (FS) intraperitoneally via Tenckhoff catheter as application interval established in the pilot for 5 days. Group 2 will receive the same dose of ceftriaxone intravenously every 24 hours and 2 liters of SF intraperitoneally at the same interval used in group 1, for 5 days. Animals of both groups will be monitored clinically and biochemically through assessments hematologic (blood count, fibrinogen, liver and kidney function and drug concentration using the technique of high performance liquid chromatography-HPLC) and peritoneal fluid (macroscopic, physical chemistry and drug concentration by HPLC). Laparoscopic and ultrasound evaluation will be applied in order to assess the drug peritoneal aggression potential. It is expected that the proposed use of the drug results in higher and durable peritoneal concentrations, and non- peritoneal reactivity. Being intraperitoneal route of application used as an adjuvant intraperitoneal treatment of peritonitis and Tenckhoff's catheter use safe and feasible, amplifying the possibilities of intraperitoneal route using.
