Genotoxic evaluation of newborn' osteoblasts of alcoholics rats

Abstract

Alcohol has teratogenic effects and its use during pregnancy can cause several changes in the fetus, including interfering in their bone development. The purpose of this study is to evaluate the genotoxic effect of alcohol 20% chronic consumption in osteoblasts obtained from the newborn rats whose mothers are treated with alcohol before and during pregnancy. For that purpose, 30 pregnant rats will be used, divided in groups according to the diet: 10 will receive alcohol 20%, 10 will belong to the isocaloric group, corresponding to the calories ingested by the group of alcohol and 10 animals will be treated with water and chow ad libitum. The rats will be conditioned to alcoholism, and for that, they will be treated from one and a half month old. Initially they will go through an adjustment period of four weeks and then will be treated daily with 20% alcohol solution until four and a half months old. After this time, they will be mated and treated throughout the gestational period. At three days of life, the newborns will be euthanized so as to remove the calvaria and start the cell culture procedures. The osteoblastic lineage cells will be isolated by sequential enzymatic digestion and the osteoblasts will be cultivated for periods of 14 days or less. Culture plates testing will take place to evaluate the genotoxic effects of alcohol based on the presence of micronuclei and comet test. The numerical data will be analyzed statistically using the ANOVA test, with significance level of 5%.
