Effect of cigarette smoke in the hippocampus of mice in a model of systemic inflammation

Abstract

Smoking is one of the major cause of chronic diseases . A previous study from our group demonstrated that cigarette smoke induces changes in antioxidant enzymes and lipid peroxidation in central nervous system ( CNS ) of mice . Also, a recently published study has shown that exposure to sixty days to the cigarette smoke increases the transcription of pro-inflammatory genes such as IL- 6, IL- 1² , TNF- ± . Therefore , this project aims to clarify the effects of exposure to cigarette smoke for fifteen consecutive days in a process of systemic inflammation induced by LPS . For this, C57BL / 6 mice are exposed to smoke a mixture of central and lateral twice a day (8h and 16h ) for fifteen days. On the last day ( 8h ) , they will be challenged with LPS iv . ( 0.1 mg / animal) or saline , forming CO groups - control , FU - cigarette smoke , LPS - LPS challenge , FPS - cigarette smoke and LPS challenge , COAG - single exposure and control FUAg - only exposure to cigarette smoke . The hippocampus samples are prepared for performing the quantitation of Western blot iNOS and IL -10 and TNF- ± by ELISA. To better understand the time course of the inflammatory process caused by cigarette smoke in the CNS , in the presence and / or absence of inflammatory stimulus the assessments will be conducted in the periods acute, of two, four and six hours after LPS challenge.
