Effect of cyclosporine on the integration of deproteinized bovine bone in a rat calvarial bone defect model

Abstract

The objective of this study will be to evaluate the effect of the cyclosporine consumption on the desproteinized bovine grafts in critical size defects in rats. Fifty-six rat will be use in this study. The animals will be randomly divide into two groups with 28 animals in each group. The animals will be allocate according to the type of drug used: Group I: animals will be subject to daily consumption of saline by gavage; Group II: animals should be subject to daily administration of Cyclosporin Csa 10mg/kg/day applied by gavage. The drugs will be appliy daily beginning 7 days before the confection of the calvarias defects until the day of sacrifice. After the confection of critical size defects in animals calvarias (5mm diameter), they will be allocate into subgroups with 14 animals each, according to the type of biomaterial that will be insert into the defect: Subgroup A: defect will be fill with blood clot; subgroup B: Defect will be fill with deproteinized bovine bone. The animals will be euthanize after 15 and 60 days of the surgeries (7 animals per period). Histomorphometric and tomographic analysis will be perform for the assessment of the volume of tissues that will fill the defect (bone, biomaterial and connective dense tissue). Regarding the statistical analysis, if the data generate by the analysis will be normal, the parametric tests of ANOVA complemented by Tukey's test will be use for inferential analysis of the data. Otherwise, the nonparametric Kruskal-Wallis complemented by Dunn's test will be use for data analysis. All tests of this study will be apply with a significance level of 95%.
