Abstract

The search for reliable and effective methods for the development and quality control of chiral drugs is extremely important for the pharmaceutical industry. In this context, two NMR techniques can contribute significantly: DOSY (Diffusion Ordered Spectroscopy) and Pure Shift (Homonuclear Broadband Decoupling). The DOSY technique is based on small differences of diffusion coefficients (D) among molecular species present in mixtures. When there is overlap of signals, due to compounds with similar D, able substances of interacting with analyte and modify their D, can be added in the sample solution. Thus, even individual enantiomers can be observed. And the Pure Shift technique may allow viewing of the enantiomers signals by the resolution gain obtained from homonuclear decoupling. Then, the aim of this project is to develop DOSY methods for qualitative analysis of chiral drugs, while methods pure shift unidimensional will be developed to enable quantification of the enantiomeric active principles. The few studies in the literature show very promising results, which demonstrate the potential of both techniques for this type of application and evidence the need for further researches in the area.
