Effect of systemic treatment with resveratrol on the progression of experimental periodontitis and arthritis

Abstract

The aim of this study is to evaluate the effect of systemic treatment with resveratrol on the progression of periodontitis in animals with rheumatoid arthritis (RA). Moreover, it also investigated the impact of resveratrol in controlling RA in animals with induced periodontitis. For this, 24 male and adult rats (Rattus norvegicus, Albinus, Wistar) will be randomly divided into the following groups: 1 - experimental arthritis + experimental periodontitis + placebo (RA + PE + PL), and 2 - experimental arthritis + experimental periodontitis + + resveratrol (RA + PE + RSV). Rheumatoid arthritis will be induced by two immunizations collagen type II and two applications of Freund's complete adjuvant (CFA). The first dose of collagen will be administered on day -21 and second dose on day -14. On day -7 two applications of CFA will be realized. On day 0, the PE will be induced by placing a ligature of silk thread in the groove of the mandibular first molars. The animals receive treatment from day 0 to day 30 by gavage and will be sacrificed 30 days after induction of PE. Weekly tests (evaluation of paw edema and analgesic test) will be realized until the end of the experiment to evaluate the effect of Resveratrol on RA. After this period, the animals will be sacrificed and the specimens will be processed for morphometric analysis of alveolar bone loss. The gum tissue will be collected for quantification of IL -17 by Luminex/MAGpix assay. Data will be analyzed statistically using the Student t-test or Mean Whitney, after the normality test (± = 5 %).
