Analysis of the expression of transcription factors Pod-1, SF-1, PINK1 and BUB1B in human adrenocortical tumors

Abstract

Pod-1/Tcf21 is expressed at sites of epithelial-mesenchymal interactions during development of different organs. Pod-1 can inhibit the transcription factor Sf-1/NR5A1 during the development of the gonads. While inhibition of Sf-1 can lead to alterations in the development of adrenal, its increased expression is associated with increased proliferation and tumorigenesis, especially in pediatric tumors. In a previous study, we showed that overexpression of Pod-1 inhibits the expression of endogenous SF-1 tumor cells of mice and human cells, by binding to E-box site of the Sf-1 promoter. Furthermore, overexpression of Pod-1 resulted in decreased StAR steroidogenic enzyme. Although it did not affect cell viability, analysis POD-1/TCF-21 in 33 Adrenocortical Carcinomas (ACC) showed an inverse relationship between the gene expression of different genes and cell cycle. The BUB1B and PINK1 genes expression are cell cycle genes considered molecular markers for prognosis of adult adrenal. Therefore, in view of the importance of SF-1 in adrenocortical tumors, especially in pediatric patients, due to the evidence that POD-1 can regulate the expression of SF-1 gene and other cell cycle genes, we hypothesized that the POD-1 expression may have value in the differential diagnosis of adrenal masses and prognosis for ACC. In this project we aim to analyze the expression of POD-1 and SF-1 in cDNA samples of 131 patient tumors, 27 pediatric tumors and 104 adults, by using TaqMan quantitative RT-PCR. The POD-1 expression will be correlated with the expression of SF-1 and the clinical and pathological outcomes of patients. The BUB1B and PINK gene expression also will be analyzed in the tumors cohort. (AU)
