Synthesis, characterization and evaluation of the capacity to inhibit topoisomerase II enzime of the Pd(II) complexes bearing thiosemicarbazone

Abstract

This project aims the synthesis , characterization of new complexes of general formula [ PdX ( TSC ) ( PPh3 ) ] TSC = { ( 2E ) -2 - [ ( 2E ) - 3 - phenyl - 1 - ylidene ] hydrazinecarbothioamide ; 2E- N-methyl- 2-[( 3 (E )-4- phenyl-3- butene -2-ylidene ] hydrazinecarbothioamide , X = Cl, I, SCN, SeCN, NNN } and evaluation of the inhibitory capacity against topoisomerase II±. The interaction between the compounds and topoisomerase will be evaluated by agarose gel electrophoresis assay. The biological activity of the most promising compounds will be evaluated by MTT assays against tumor and normal cells .The compound 2E- N-methyl- 2-[( 3 (E )-4- phenyl-3- butene -2-ylidene ] hydrazinecarbothioamide will be synthesized by the reaction between benzylideneacetone and 4-methyl -3- thiosemicarbazide. The compounds will be characterized by elemental analysis, IR and NMR spectroscopies, ESI/MS mass spectrometry, thermogravimetry (TG) and single crystal X-ray diffraction methods.
