Abstract

This project aims to develop selenium- tellurium-coumarin derivatives for studies of suppression and fluorescence emission and biological applications. Certain diseases or disorders are associated to oxidative stress (Reactive Oxygen Species, ROS) and both Te and Se atoms are susceptible to oxidation by such species. Due to the heavy atom effect both elements, when connected directly to fluorophoric cores are capable of inhibiting the fluorescence emission. If these elements are oxidized, the system becomes fluorescent again. Thus, we plan the preparation of coumarin derivatives containing Se and Te atoms for cell labeling studies in which endogenous oxidants are overexpressed. These studies are conducted with simulated physiological solutions in which the production of the oxidant is overexpressed. This proposal is based on recently collaboration with a research group from the Universidade Nova of Lisboa, which has extensive experience in the fluorescent sensors research area. In this aspect we should also mention that for the feasibility of experimental studies involving both teams, a bilateral project was recently approved and funded by CNPq through the Science Without Borders Program in which 3 post-doctoral scholarships were granted to members of the Portuguese team to come and conduct the experiments in our group and 3 PhD scholarships for sandwich students of our group.
