Abstract

Bone resorption is a process that involves a series of steps, starting with the proliferation of precursor of osteoclasts from hematopoietic lineage monocytes, followed by cellular differentiation and maturation, resulting in the degradation of organic collagen and inorganic matrices of the bone tissue. The specific objective of this research is to investigate the mechanisms involved in regulating the synthesis of leukotrienes and their role in the differentiation of osteoclasts from hematopoietic lineage cells of mice (J774). Initially the cells will be stimulated with immunostimulants and then, with the purpose of investigating the role of 5-Lipoxygenase pathway activation, pharmacological inhibition of via with the pharmacological inhibitor MK-886 will be performed. The mRNA expression will be evaluated by means of RT-PCR in real time. Statistical analysis will be carried out according to the distribution of data using Kruskal-Wallis or ANOVA tests (± = 0.05), according to the distribution of data.
