Abstract

Diabetes mellitus during pregnancy is associated with higher levels of urinary incontinence and pelvic floor muscle dysfunction, because the abdominal muscles take part on the urinary continence mechanism , this project will analyze the rectus abdominis muscle of pregnant rats with severe diabetes (blood glucose levels higher than 300 mg / dL) and moderate (blood glucose levels between 120 and 300 mg / dL). Wistar rats were divided into six groups (n = 13 animals / group): non-diabetic and non-pregnant (control), non-diabetic pregnant, moderate diabetic non-pregnant, severe diabetic non-pregnant, moderate diabetic pregnant and severe diabetic pregnant. Moderate diabetes was induced on day 1 of birth by subcutaneous administration of streptozotocin at a dose of 100 mg / kg of body weight diluted in 0.1 mol / l citrate buffer (pH 4.5). On the inclusion criterion was blood glucose levels higher than 400 mg / dL in day 5 of life. Severe diabetes was induced in adulthood (90 days of life) by intraperitoneal administration of streptozotocin at a dose of 40 mg / kg body weight. The inclusion criterion was blood glucose levels higher than 300mg/dL. At the end of the experiment, the rats were anesthetized, euthanized and the rectus abdominis muscle was frozen in liquid nitrogen and stored at -80 ° C and subjected to cryostat sections of 10 µm thick.. The sections were submitted to cytochemical staining for muscle and extracellular matrix (colagen). Thus, after the removal and processing and staining of the muscle, we intend to analyze the structure of the extracellular matrix of these rats in partnership with the "Case Western Reserve University". The area occupied by connective tissue around the muscle fibers of the rectus abdominis will be analyzed. Furthermore, to understand the structural effects of diabetes and pregnancy interaction on the extracellular matrix and on the rectus abdominis, and verify changes in different glycemic intensities.
