Abstract

Bothrops venom contains several major components including metalloproteinases, serine proteinases, phospholipase A2 and lectins. Among these components, metalloproteinases (SVMPs) are the most abundant in most Bothrops venoms reaching 38% of the B. neuwiedi pauloensis venom total components. Several conditions such as phylogeny, ontogeny, sex, environment or diet can alter the venom composition, mainly on the SVMPs type and abundance. The mechanism of action of SVMPs is related to the proteolysis of extracellular matrix components, plasma proteins and cell surface proteins. In our Master's project, we identified the presence of several SVMPs in the B. neuwiedi venom. In addition to structural differences, these toxins acted on different targets of mammal hemostatic system. For example, two class P-I have been isolated to homogeneity and their partial amino acid sequences (40% coverage) were identical, although they presented distinct pI and functions. Our goal in this project is to elucidate the complete amino acid sequences of these two SVMPs and their three dimensional structure in order to understand the functional differences between these enzymes.
