Evaluation of biofilm formation on titanium discs with micro and nanotopography: an in situ study

Abstract

Changes in the surfaces of implants have been performed in order to improve their osseointegration and integration with the soft tissues. These changes may alter the topography, free energy, wettability, chemical composition, characteristics that may in turn influence both on cell adhesion as well as bacterial adhesion. Recently, nanotopography were evaluated for the ability to osseointegration, showing better results in relation to microtopography and machined titanium surphaces. Thus, the present study aims to evaluate the influence of nanotopography of titanium surfaces in microbial adhesion process, in situ. Titanium discs were produced and subjected to various methods for obtaining surfaces; machined, micro and nanotopography. The topography, morphology, and chemical composition of these discs have been characterized in a previous study, showing that the roughness of the disk with microtopography is greater than that of the nanotopography disc. The experimental groups are: machined discs (n = 30), microtopography (n = 30) and nanotopography (n = 30). For the in situ evaluation, individual trays will be made and each will set six bodies-specimens, two in each group. Fifteen volunteers will use the trays containing the titanium discs for 24 h. After this period, the discs will be evaluated by the method of microbial culture for subsequent total count of aerobic and anaerobic bacteria. In addition, samples will be evaluated by real - time PCR for total bacterial quantification and specific quantification of Streptococcus oralis and Streptococcus mitis.
