In vivo study of the anti-inflammatory active of Otosporin in rat's teeth submitted to dental bleaching: histological and immunohistochemistry analysis

Abstract

The Otosporin is a drug indicated for the treatment of external otitis, and also employed in endodontic procedures such as pulp capping, pulpotomy and as a root canal dressing in cases of biopulpectomy. It's job becomes effective for the high penetration capability of this product. Although widely studied by medical and dental field has not yet been explored as dental post-bleaching agent. The aim of this study is to evaluate the anti-inflammatory active of Otosporin in teeth submitted to dental bleaching. 40 rats will be divided into 4 groups of 10 animals each. The animals will be anesthetized and the dental bleaching will be held in 3 right and left upper molars using 35% hydrogen peroxide following the protocols: G1A - bleaching agent followed by Otosporin; G2A - bleaching agent followed by Otosporin vehicle; G1B - bleaching agent vehicle followed by Otosporin; G2B - bleaching agent vehicle followed by Otosporin vehicle. After 2 days, the animals will be sacrificed and the jaws removed and processed for laboratory evaluation in optical microscopy. The tissue sections will be stained with HE and marked by immunohistochemistry for pro-inflammatory cytokine TNF-±, and evaluated by pre-established criteria by assigning scores to microscopic events. The results will be analyzed statistically by the Kruskal Wallis and Dunn tests with significance level of 5 % (p < 0.05).
