Abstract

The keloids is a dermal and fibrous lesion that grows beyond the original scar margin for a long period of time; usually does not regress spontaneously and may have relapsed after treatment. Appears mainly after trauma to the skin by various agents , such as after placement earring or pierced ears , in the abdomen appears after hysterectomy or Caesarean section, and in the chest after mastectomy or heart surgery. There is still not an appropriate treatment for keloids , moreover this disease affects quality of life , for that reason knowledge of its formation and molecular and genetic characteristics are só necessary. Recent studies claim that the keloid is a multifactorial disorder involving nutritional , biochemical, immunological and genetic factors that play a role in abnormal healing process ,more the action of cytokines and growth factors that are involved in inflammatory and healing processes of formation of keloid , as well as radioactive oxygen species (ROS) that are causing apoptosis and senescence , and would cause that the cells do not respond appropriately to chemical signals, stimulation of growth by changing the cytological , biochemical and genetic characteristics. Thus , this study aims to know the gene expression of 84 genes markers of oxidative stress by method of qPCR array in keloids . The possibility of using this method in patients with keloids in specific areas such as the ear lobe , naïve compared to the control group was drawn . This method aims to construct a profile of genes expressed in each group of patients , build networks of expressed genes , guiding potential clinical interventions and enable the development of future studies to allow for better prospects in the clinical treatment of patients with keloid. Methods: 10 patients with keloids will be included in the earlobe region that conform the keloid group and 5 healthy patients without keloids conform to the control group . Therefore, the genetic material is extracted from samples and will be made a gene pool of each group that will be analyzed 84 genes markers of oxidative stress using PAHS - 065Z , Profiles RT PCR Array Human Oxidative Stress kit. The experiments will be performed in triplicate.
